108 实验动物与比较医学LaboratoryAnimal andComparativeMedicine Apr2011.31(2) .doi:10.3969/j.issn.1674—5817.2011.02.008 水溶性氮酮对斑马鱼胚胎药物 渗透及致畸性作用的影响 陈锡强 ,韩利文.-2,刘可春 ,侯海荣 ,王希敏 ,何秋霞 (1.山东省科学院生物研究所,济南250014;2.天津中医药大学,天津300193) 【摘要]目的 研究水溶性氮酮对白藜芦醇透过斑马鱼胚胎性能与斑马鱼致畸性的影响。方法 利 用对斑马鱼胚胎发育致畸作用,考察氮酮在斑马鱼筛选模型上的适用性,确定氮酮的使用剂量;利 用氮酮的透皮促进作用,观察对白藜芦醇抑制斑马鱼节间血流的影响。结果在0.01~0.5 gg/ml范 围内氮酮对斑马鱼较安全,0.5 gg/ml溶性氮酮可显著促进斑马鱼对白藜芦醇的透过,斑马鱼节间 血管的抑制率达到了22.9%。结论 浓度高于0.5 gg/ml的水溶性氮酮对斑马鱼胚胎的发育有明 显致畸作用;0.5 gg/ml的氮酮对白藜芦醇的斑马鱼胚胎透过效果最佳。 【关键词】氮酮;白藜芦醇;透皮吸收;斑马鱼 [中图分类号】Q95—33 [文献标识码]A [文章编号]1674—5817(201 1)02—0108.03 斑马鱼(Danio rerio)是一种热带鱼,因体侧有 银蓝色的纵向条纹而得名。斑马鱼胚胎发育过程中 血管生成过程与肿瘤血管生成模式相近,因此被认 为是一种较理想的筛选抑制血管生成类抗肿瘤药物 的活体动物模型。在用于血管抑制药物筛选时, 1材料与方法 1.1实验动物 选用了荧光蛋白转基因斑马鱼。转基因斑马 鱼(TG)种鱼来自于哈佛大学医学院,实验所用的 斑马鱼为血管荧光转基因斑马鱼(VEGFR2:GFP)。 斑马鱼的养殖和培育参照westerfie.1 A[21的方法。选 施药方式简单,将待筛样品溶于有机溶媒或水中, 通过扩散或吸收作用进入胚胎发挥作用。然而药物 的极性、溶解性差异较大,故有必要考察药物对 斑马鱼胚胎的透过性能。白藜芦醇又名虎杖甙元, 属葸醌萜类化合物,是一种天然的抗肿瘤、抗心 血管疾病、抗突变、抗菌、抗炎、抗氧化、诱 导细胞凋亡及雌激素调节物质。研究表明白藜芦醇 具有抑制血管形成的作用[zl。氮酮是药物制剂研究 择成熟斑马鱼雌雄一对,傍晚时放入培育缸中,产卵 后取出受精卵,单独放入28℃光照培养箱备用。 1.2主要试剂和仪器 白藜芦醇购自上海同田生物技术公司,分子式 C H :0s,纯度≥98%,置冰箱中备用。水溶性氮酮 购自山西芮城东泰中间体有限公司,纯度≥98.5%。 中广泛应用的一类促渗剂,本实验对氮酮在斑马鱼 体系中对药物的促渗作用进行考察。 白藜芦醇样品配制成1 mg/ml冰箱备用,氮酮用纯 水配制为成终浓度100、10、1、0.1、0.0l gg/ml 水溶液备用。主要实验仪器:COIC XSZ—HY荧光显 微镜和COIC倒置生物显微镜XDS一1B,恒温培养箱。 【收稿日期】2010.07—13 【基金项目]山东省科学院博士基金资助项目(NoSY2007BS10) [作者简介】陈锡强(1972一),男,博士,研究方向:天然产物抗肿 瘤筛选。E—mail:cxqq一1972。163.COITI 【通讯作者]刘可春(1964.),男,博士,研究员,研究方向:天然药 物活性筛选,E—mai1:hliukcha。keylab.net 1.3氮酮促进白藜芦醇透皮实验 鱼胚胎于24 hpf后取出,脱去膜胚胎加入96 孔板中;将试验药物加入96孔板中,分别设立正常 对照组、氮酮组、白藜芦醇组、氮酮加白藜芦醇 1组和氮酮加白藜芦醇2组各1O只胚胎;生理组不 11O 实验动物与比较医学Laboratory Animal and Comparative Medicine 3讨论 斑马鱼是一种较理想的筛选抑制血管生成类抗 肿瘤药物的活体动物模型【 ,其躯干部体节问的血管 开始由背部的动脉出芽形成于相邻体节之间,此过 程与肿瘤增长新血管形成类似,初期胚胎透明易于在 显微镜下观察『4】。利用荧光蛋白标记VEGFR一2的斑 马鱼是抑制血管形成药物的筛选和评价是理想动物 模型【51引。作者发现部分抑制血管生成化合物对斑 马鱼胚胎的侧枝血管的抑制并不明显,考虑是否与 药物的斑马鱼胚胎透过能力有关。这个因素与斑马 鱼模型的特点有关,作者在利用中药提取物抑制血 管形成筛选的过程中,处于斑马鱼胚发育的前72 h, 鱼胚属于闭口期【7],所以药物无法通过消化道吸收 进入鱼体,具备透皮能力的药物且具备抑制血管形 成作用的药物才能显示出活性,而对透皮作用很差 的药物,这样的样品往往会按照无抑制血管生成活 性来处理。 研究发现1.37 ̄22.8 gg/ml白藜芦醇有抑制牛 主动脉内皮细胞迁移的作用,0.23~0.57 gg/ml白藜 芦醇显著抑制人脐静脉内皮细胞迁移,其阻滞血管 形成的作用途径是通过刺激csk家族激酶高表达抑 制src激酶的活性,使得钙粘附蛋白磷酸化减低, 从而使内皮细胞管形成无法实现[s, 。作者实验中 发现,作为传统的透皮剂的水溶性都不佳,使得无 法控制透皮剂浓度。常用的药物辅料氮酮具可改变 脂质双分子层的致密性,增加脂质流动性,且对细胞 内的蛋白质无影响、毒性及刺激性小等优点 ,1”, 因此,本实验将改性后的水溶性氮酮作为渗透剂的 首选,放弃了丙二醇和薄荷油等常用透皮剂。从 本实验结果可以看出,白藜芦醇存在透皮障碍,单独 作用的白藜芦醇未显示对节间血管的抑制作用;随着 透皮剂氮酮剂量的加大,白藜芦醇的抑制血管形成 作用也呈现增加的趋势,说明氮酮透皮作用对白藜 芦醇渗透有协助作用;但氮酮浓度增加会引起斑马鱼 胚胎的畸形率上升,实验中作者发现0.5 ml氮酮 的促透皮作用最佳,且对鱼体发育影响较小,同 时可以满足药物促渗透筛选的要求。 有研究认为氮酮促渗可能受药物的分配参数、 细胞膜的渗透性、磷酸化等因素相互影响【12]。氮 酮促进白藜芦醇透皮的作用途径值得探讨,白藜芦 醇是属于葸醌甙元,氮酮对此类天然产物具有促进 渗透作用还需要进一步验证。 [参考文献】 [1]ThaloorD,SinghAK,SidhuGS,eta1.Inhibiitonofangiogenic differentiation of human umbilical vein endothelial cells by curcumin[J].Cell Growth Differ,1998,9(4):305—312.
